As CAR-T therapies continue to advance, the use of VHH in CAR design has become increasingly common. This antibody demonstrates excellent binding activity to Jurkat-CAR cells based on VHH nanobodies, and is recommended for use in CAR-T cell validation and identification.

In this experiment, primary T cells were used as CAR-negative controls. A mixture containing 9.43% CAR-positive T cells and primary T cells served as the positive sample for separation. Magnetic separation of CAR-T cells was performed using the biotin-labeled MonoRab™ Rabbit Anti-Camelid VHH Cocktail [Biotin] (GenScript, A02015) together with anti-biotin magnetic beads. The separated cells were analyzed by flow cytometry to evaluate the separation efficiency. As shown in the figure, the proportion of CAR-positive cells increased from 9.43% in the pre-mixed sample to 88.9% after magnetic separation, demonstrating that this antibody is well-suited for CAR-T cell magnetic separation.
Sandwich ELISA Application

Standard curve of VHH Sandwich ELISA. The VHH Sandwich ELISA assay is developed by using MonoRab™ Rabbit Anti-Camelid VHH Antibody, mAb (GenScript, A01860) and MonoRab™ Rabbit Anti-Camelid VHH Cocktail [HRP] (GenScript, A02016) as the capture and detection antibodies, respectively.

In this ELISA assay, the detection antibody was labeled with horseradish peroxidase (HRP), if you needs an unconjugated antibody you can choose MonoRab™ Rabbit Anti-Camelid VHH Cocktail (GenScript, A02014). GenScript can provide customized conjugation services for this product per the customer's request.

The sensitivity of detecting VHH is up to 39 pg/ml.
