small molecule library

A collection of related structures synthesized for screening for a desired property. The library may by synthesized by parallel, split or spatially addressable synthesis. parallel synthesis is a single-batch method that uses a mixture of reagents at each step of the synthesis to generate the diversity of products. split synthesis requires a solid-phase support to allow separation of products at each synthetic step. For example, a polymer may be assembled by coupling monomers A, B and C to beads; the separate batches are filtered and combined, then divided again into thirds for coupling with monomers D, E and F. After n cycles, there will be 3n sequences present. spatially addressable synthesis localizes the reactions on specific locations on a chip, a solid surface ruled into a grid, each locus on which receives a unique sequence of reagents. The products of each synthetic method are exposed to a selective procedure. The position on a chip directly identitfies the structure assembled there. If a binding property is sought, localization to a ligand on a solid-phase surface will assist selection from a library of dissolved structures. Coding of beads during their synthesis allows their identification after selection; such coding may be the parallel assembly of a protein or nucleic acid during each step of the synthesis, polymers of which are easily sequenced by available technologies.Borman, S.
