Description

PiggyBac is a DNA transposon and a promising alternative to Sleeping Beauty. Piggybac is originally isolated from the cabbage looper moth Trichoplusia ni genome, is distinguished by its ability to excise precisely. (Zheng Zhang, et al, J Comput Biol 2000; 7(1-2):203-14.)

This mRNA features:
- A Cap 1 structure with high capping efficiency
- 100% substitution with N1-methyl-pseudouridine (m1Ψ) for improved protein expression and reduced innate immune response
- GenScript’s patented UTR to enhance translation
- A 100A poly(A) tail to mimic natural mature mRNA

Applications
Seed 5 × 10⁴ HEK293T cells per well in a 24-well plate one day before transfection. On the day of transfection, replace the culture medium with OptiMEM supplemented with 10% FBS. Prepare a transfection mix containing piggyBac mRNA and donor DNA (recommended molar ratio 1:2, total amount 1μg), and deliver using Lipofectamine 2000 (or equivalent). After transfection, passage cells on Day 3, 6, 10, and 12. Collect samples at each time point for flow cytometry analysis.

For laboratory research use only. Direct human use, including taking orally and injection and clinical use are forbidden.
