Seed 5 × 10⁴ HEK293T cells per well in a 24-well plate one day before transfection. On the day of transfection, replace the culture medium with OptiMEM supplemented with 10% FBS. Prepare a transfection mix containing piggyBac mRNA and donor DNA (recommended molar ratio 1:2, total amount 1μg), and deliver using Lipofectamine 2000 (or equivalent). After transfection, passage cells on Day 3, 6, 10, and 12. Collect samples at each time point for flow cytometry analysis. |
