Two-Step DNA Mutant Library Strategy to Achieve a 1000-Affinity Boost
Through two rounds of optimization using GenScript’s DNA Mutant Library, significant improvements were achieved in the affinity maturation of a monoclonal antibody. Initially, a saturation scanning library was created with 63 amino acids and 19 non-wild type amino acids per position across 6 CDR regions. The top 5 candidates were selected for a second round of combinatorial optimization. The novel antibody from this mutant library showed a 1000-fold increase in binding affinity compared to the wildtype.
